Sirtuins and NAD+

Prof. David Sinclair has conducted extensive research on sirtuins, a group of enzymes that play a key role in regulating the aging process. Sirtuins are activated by NAD+ (nicotinamide adenine dinucleotide), a molecule whose levels in the body decrease with age. By increasing NAD+ levels, sirtuins can be activated, leading to improved DNA repair, increased energy production in the mitochondria and an overall improvement in cellular function[1][3].

Epigenetic changes

Sinclair has also studied the role of epigenetic changes in the aging process. Epigenetic factors such as SIRT1 can be redistributed in response to DNA damage, leading to aging of cells. Through targeted epigenetic modulation, these changes can potentially be reversed, allowing cells to rejuvenate[1].

Xenohormesis Another interesting concept is the xenohormesis hypothesis, which states that plant compounds produced under stress conditions can activate longevity mechanisms in animals and humans. Resveratrol, a polyphenol from grapes, is a well-known example of such a substance that activates SIRT1 and can thus extend lifespan[1][6].

NMN and NR

Nicotinamide mononucleotide (NMN) and nicotinamide riboside (NR) are precursors of NAD+ and have been shown in studies to increase NAD+ levels and thus reduce age-related loss of function in cells. These substances are already available as dietary supplements and are being intensively researched to confirm their effectiveness and safety in humans[3][6].
